Fay's Drug began in 1958, when Henry A. Panasci Jr. and his father opened the first store in Fairmount, New York. It was named for Faye Panasci, Henry Jr.'s wife; the final e was omitted to reduce sign costs. Incorporated as Fay's Drugs Company, Inc. on October 20, 1966, the business established headquarters in Syracuse, with much of its operations in Liverpool. It went public in 1969 and entered Rochester in 1971, later acquiring the Key Drug chain there in 1979. That year it also bought the Mann and Crafts chains in North and South Carolina, but sold those stores in 1981. Fay's acquired Carls Drugs in 1991 and Peterson's 12-store Western New York chain in 1994. At its peak, the company operated more than 300 stores in New York and Northeast Pennsylvania, alongside Wheels Discount Auto Supply and The Paper Cutter. It sold those non-drug businesses in the 1990s. In 1996, JCPenney bought Fay's for $285 million in stock; New York stores kept the Fay's name, while stores elsewhere became Thrift Drug.
